I want to do a battery load test with volt meter.  Which fuse would be best to pull in your opinion.  Or what is best.

I didn't think you needed to pull a fuse to do the test?  Are you trying to prevent it from starting?  If so just disconnect the vp44 plug.

If your doing voltage test off batteries you want to just disconnect the battery grounds. This way the batteries are isolated.
